1.Backup/Incremental backup on Local Computer(VM Explorer is installed on the local computer): the data runs from the ESX/ESXi server to the local computer

2.Backup/Incremental backup on Local Computer(mapped network drive): the data runs from the ESX/ESXi server to the computer where VM Explorer is installed and then to the mapped network

3.Backup same host as VM: the data runs from/to the same server because source server and target server are the same.  If you enable VD Service the backup will be automatically executed using the host so that you will copy directly the disks of the VM without the need to any outside communication.

4.Backup between ESX/ESXi with agent enabled: the data runs from host to host because they use the agent connection between servers

5.Backup between ESX/ESXi without agent enabled: the data runs from the source server to the local machine where VMExplorer is installed through the VM Explorer service and then from the local machine to the target server

The condition for the replication is the same as number 3 and number 4.
The only exception is for incremental replication where data runs from the source server to the local machine where VMExplorer is installed and than from the local machine to the target server.
