Currently, you are not able to do this in VigilEnt Security Manager (VSM).  The action 'Assign Registry Value' only allows you to make a change to an existing registry value.  An enhancement request has been submitted for this feature.  It will be considered for a future release of the VigilEnt Security Agent for Windows and VSM.
