There should be no interruption of service to the Directory and Resource Administrator (DRA) server during a NT backup. 

Tech Support set up a test to test the effect of a backup job and DRA.  A native NT backup process was setup and run during the time period of all three types of cache refresh processes (accounts, domain and resource), a last logon statistics gathering job and an import of the DRA Reporting tool.  During all test, the backup process was never interrupted and it completed without error as well as the DRA server operations were not interrupted.
