When I migrate my distribution lists from my source organization to my target organization, will I see the changes in the memberships from both sides if I run directory synchronization (EMCLI DIRSYNC)?

After you initialize directory synchronization (EMCLI DIRSYNC), additions made to distribution lists in either source or target organization are copied, so that changes are reflected in both organizations. EMCLI DIRSYNC ensures the Global Address List (GAL) and mail flow between the organizations is up to date and not interrupted.  This allows you to operate your Exchange organizations in parallel during your migration.

You will need to run EMCLI DIRSYNC to replicate these changes in memberships.  To see these changes on both the target and the source, regardless of where you make the change in membership, you will most likely need to run EMCLI DIRSYNC with the '/twoway' switch.
